Abstract
Dermatofitosis merupakan infeksi jamur superfisial yang menyerang jaringan keratin seperti kulit, rambut, dan kuku. Infeksi ini disebabkan oleh kelompok jamur dermatofita, salah satunya adalah Trichophyton rubrum yang merupakan penyebab paling sering dermatofitosis pada manusia. Pertumbuhan jamur dermatofita dipengaruhi oleh berbagai faktor lingkungan, salah satunya adalah suhu. Variasi temperatur dapat mempengaruhi laju pertumbuhan, viabilitas, serta karakteristik morfologi koloni jamur. Penelitian ini bertujuan untuk mengetahui efek variasi temperatur terhadap pertumbuhan T. rubrum isolat Jakarta pada media Sabouraud Dextrose Agar (SDA). Penelitian ini merupakan penelitian eksperimental yang dilakukan di laboratorium Parasitologi FK UKI. Isolat T. rubrum diinokulasi pada media SDA dan diinkubasi pada empat variasi suhu, yaitu suhu kamar (±28°C), 37°C, 4°C, dan -20°C dengan tiga kali replikasi. Pengamatan dilakukan secara makroskopis pada hari ke-3, hari ke-7, dan hari ke-14 untuk menilai waktu munculnya koloni, warna permukaan koloni, serta perubahan warna reverse. Pada hari ke-14 dilakukan pengamatan mikroskopis menggunakan pewarnaan Lactophenol Cotton Blue (LPCB) untuk mengamati struktur hifa dan mikrokonidia. Hasil penelitian menunjukkan pertumbuhan koloni T. rubrum mulai tampak pada hari ke-3 pada suhu 37°C dan suhu kamar (±28°C), dengan pertumbuhan koloni lebih cepat dan lebih tebal pada suhu 37°C. Pada suhu 4°C dan suhu -20°C tidak ditemukan pertumbuhan koloni selama masa inkubasi. Namun, setelah dipindahkan kembali ke suhu kamar koloni dapat tumbuh dengan baik. Secara mikroskopis seluruh preparat menunjukkan gambaran hifa bersekat dan mikrokonidia berbentuk seperti tetesan air mata yang merupakan karakteristik T. rubrum. Berdasarkan hasil penelitian ini, disimpulkan variasi temperatur mempengaruhi pertumbuhan T. rubrum. Suhu yang mendekati suhu fisiologis manusia lebih mendukung pertumbuhan jamur, sedangkan suhu rendah dapat menghambat pertumbuhan tanpa menghilangkan viabilitas jamur./Dermatophytosis is a superficial fungal infection that affects keratinized tissues such as skin, hair, and nails. This infection is caused by a group of dermatophyte fungi, notably Trichophyton rubrum, which is the most common cause of dermatophytosis in humans. The growth of dermatophytes is influenced by various environmental factors, one of which is temperature. Temperature variation can affect the growth rate, viability, and morphological characteristics of fungal colonies. This study aimed to determine the effect of temperature variations on the growth of T. rubrum Jakarta isolate on Sabouraud Dextrose Agar (SDA) media. This study was an experimental study conducted at the Parasitology Laboratory, Faculty of Medicine, Universitas Kristen Indonesia. Trichophyton rubrum isolates were inoculated onto SDA media and incubated at four temperature variations, namely room temperature ±28°C, 37°C, 4°C, and -20°C with three replications. Observations were conducted macroscopically on day 3, day 7, and day 14 to assess the time of colony appearance, colony surface color, and changes in reverse pigmentation. On day 14, microscopic observation was performed using Lactophenol Cotton Blue (LPCB) staining to observe the structure of hyphae and microconidia. The results showed that the growth of T. rubrum colonies began to appear on day 3 at 37°C and room temperature (±28°C), with faster and thicker colony growth was observed at 37°C. At temperatures of 4°C and -20°C, no colony growth was observed during the incubation period. However, after being transferred back to room temperature, the colonies were able to grow well. Microscopically, all preparations showed septate hyphae and tear-shaped microconidia, which are characteristic features of T. rubrum. Based on the results of this study, it can be concluded that temperature variation affects the growth of T. rubrum. Temperature close to human physiological temperature support fungal growth, while low temperatures can inhibit growth without eliminating fungal viability.
