Abstract
Kulit memiliki mikroorganisme Staphylococcus sp. yang bersifat komensal dan berpotensi menjadi patogen oportunistik, yang menyebabkan infeksi pada kulit, sistem pencernaan, dan sistem pernapasan. Salah satu antibiotik yang biasa digunakan untuk mengatasi infeksi Staphylococcus aureus adalah penisilin, namun pada beberapa kasus ditemukan resistensi. Oleh karena itu, dilakukan penelitian pencarian antibakteri alternatif dengan efek samping minimal dan memiliki sensitivitas yang baik. Penelitian ini bertujuan untuk mengevaluasi aktivitas antibakteri ekstrak beras hitam varietas cempo ireng (Oryza sativa L. indica) terhadap bakteri Staphylococcus aureus. Ekstraksi beras hitam dilakukan menggunakan metode maserasi dengan pelarut etanol 96% dan kloroform 99,8%. Pengujian aktivitas antibakteri menggunakan metode difusi cakram Kirby-Bauer dengan konsentrasi ekstrak 100 mg/mL, 50 mg/mL, 25 mg/mL, 12,5 mg/mL, dan 6,25 mg/mL. Kontrol positif menggunakan antibiotik gentamisin dan DMSO sebagai kontrol negatif. Hasil penelitian menunjukkan terdapat zona hambat pada gentamisin sebesar 16,28 mm dan 16,35 mm, namun tidak terdapat zona hambat pada ekstrak beras hitam konsentrasi 100 mg/mL, 50 mg/mL, 25 mg/mL, 12,5 mg/mL, 6,25 mg/mL, dan DMSO. Dengan demikian, dapat disimpulkan bahwa ekstrak beras hitam varietas cempo ireng (Oryza sativa L. indica) tidak membentuk zona hambat terhadap Staphylococcus aureus pada konsentrasi dalam penelitian ini. / Staphylococcus sp. an opportunistic pathogens that is commensal on the skin, which can cause infections of the skin, digestive system and respiratory system. One of the antibiotics commonly used to treat Staphylococcus aureus infections is penicillin, but resistance has been observed in several cases. Therefore, this research was carried out on alternative antibacterials with minimal side effects and decent sensitivity. This study aims to assess antibacterial activity of black rice extract of the cempo ireng variety (Oryza sativa L. indica) against Staphylococcus aureus. Black rice extraction was using the maceration method with 96% ethanol and 99,8% chloroform as solvents. Antibacterial activity testing used the Kirby-Bauer disc diffusion method with extract concentrations of 100 mg/mL, 50 mg/mL, 25 mg/mL, 12.5 mg/mL, and 6.25 mg/mL. The positive control used the antibiotic gentamicin and DMSO as a negative control. The results showed that there were inhibition zones for gentamicin of 16,28 mm and 16,35 mm, but no inhibition zones containing black rice extract at concentrations of 100 mg/mL, 50 mg/mL, 25 mg/mL, 12.5 mg/mL, 6.25 mg/mL, and DMSO. Thus, it can be concluded that the black rice extract of the cempo ireng variety (Oryza sativa L. indica) did not form an inhibitory zone against Staphylococcus aureus at all concentrations in this study.
