Abstract
The accumulation of free radical causes several human diseases. Papaya seeds have been known as a potent antioxidant and also anticancer. This study evaluates the antioxidant and cytotoxicity activities of California and Bangkok varieties of papaya seeds. In this study, antioxidant activity was analyzed with DPPH (1,1diphenyl-2-picrylhydrazyl) method. The results showed that the IC50 values of Bangkok papaya seed extracts were 22.2 ppm, while the IC50 values for California papaya seed extracts were 24.4 ppm. This present study assessed the cytotoxicity effect by using the MCF-7 (breast cancer cells) and CHO (Chinese Hamster Ovarium) cells. The California papaya seed extracts have a more cytotoxic effect than Bangkok papaya seeds extract. The highest concentration of metabolite was oleic acid in Bangkok papaya seed and β-sitosterol in California papaya seed extracts. Keywords : anticancer; antioxidant; cytotoxicity; free-radical; papaya seeds. / Berbagai penyakit yang menyerang manusia banyak diakibatkan oleh terakumulasinya radikal bebas di dalam tubuh. Biji pepaya telah diketahui memiliki aktivitas antioksidan dan antikanker. Oleh karena itu, penelitian ini bertujuan untuk mengetahui aktivitas antioksidan dan efek sitotoksik terhadap sel kanker dari ekstrak biji pepaya jenis california dan bangkok. Uji aktivitas antioksidan yang dilakukan menggunakan metode DPPH (1,1-difenil-2-pikrilhidrazil), menghasilkan nilai IC50 sebesar 22.2 ppm untuk ekstrak biji pepaya bangkok dan nilai IC50 sebesar 24.4 ppm untuk ekstrak biji pepaya california. Uji sitotoksik dalam penelitian ini dilakukan terhadap sel MCF-7 (sel kanker payudara) dan sel CHO (Chinese Hamster Ovarium). Ekstrak biji pepaya california memiliki aktivitas sitotoksik yang lebih tinggi dibandingkan ekstrak biji pepaya bangkok. Kandungan metabolit tertinggi pada ekstrak biji pepaya bangkok adalah asam oleat, sedangkan ekstrak biji pepaya california adalah β-sitosterol.
