Abstract
Latar belakang: Staphylococcus aureus adalah bakteri penyebab berbagai infeksi, dari infeksi kulit hingga sistemik. Ekstrak bunga mawar merah (Rosa damascena mill) memiliki potensi antibakteri terhadap bakteri ini, berkat kandungan senyawa aktif seperti flavonoid dan minyak esensial yang merusak membran sel bakteri dan mendukung sistem imun. Tujuan penelitian: Penelitian ini bertujuan menguji efektivitas ekstrak mawar dalam menghambat pertumbuhan S. aureus. Metode penelitian: Penelitian ini menggunakan metode difusi Kirby-Bauer dengan desain post-test only control group. Sampel bakteri S. aureus diuji dengan ekstrak bunga mawar merah (Rosa damascena mill) pada konsentrasi 20%, 40%, 60%, 80%, dan 100%. Kontrol positif adalah gentamisin, dan kontrol negatif menggunakan DMSO 20%. Diameter zona hambat diukur setelah inkubasi 24 jam. Data dianalisis secara deskriptif untuk mengevaluasi efektivitas antibakteri ekstrak tersebut. Hasil penelitian: Ekstrak bunga mawar merah (Rosa damascena mill) menunjukkan aktivitas antibakteri terhadap S. aureus dengan efektivitas yang bervariasi tergantung konsentrasi. Konsentrasi 100%, 80%, dan 60% memiliki daya hambat kuat (diameter zona hambat rata-rata 12,95 mm hingga 20,75 mm). Konsentrasi 40% dan 20% menunjukkan daya hambat sedang (8,6 mm dan 6,8 mm). Kontrol positif (gentamisin) memberikan zona hambat tertinggi (20,75 mm), sedangkan kontrol negatif (DMSO 20%) tidak menunjukkan zona hambat. Kesimpulan: Melalui pengujian dengan metode Kirby-Bauer menunjukkan bahwa ekstrak kelopak bunga mawar merah pada konsentrasi 100%, 80%, 60% termasuk dalam daya hambat kuat. Sedangkan, pada konsentrasi 40% dan 20% termasuk dalam katagori daya hambat sedang. Hasilnya adalah ekstrak kelopak bunga mawar merah terbukti efektif dalam menghambat pertumbuhan bakteri S. aureus. / Background: Staphylococcus aureus is a bacterium that causes various infections, from skin infections to systemic infections. Red rose flower extract (Rosa damascena mill) has antibacterial potential, due to the content of active compounds such as flavonoids and essential oils that damage bacterial cell membranes and support the immune system. Objective: This study aims to test the effectiveness of rose extract in inhibiting the growth of S. aureus. Research method: This study used the Kirby-Bauer diffusion method with a post-test only control group design. S.aureus bacterial samples were tested with red rose flower extract (Rosa damascena mill) at concentrations of 20%, 40%, 60%, 80%, and 100%. The positive control was gentamicin, and the negative control used DMSO 20%. The diameter of the inhibition zone was measured after 24 hours of incubation. The data were described descriptively for the antibacterial effectiveness of the extract. Results: Red rose flower extract (Rosa damascena mill) showed antibacterial activity against S. aureus with varying effectiveness depending on the concentration. Concentrations of 100%, 80%, and 60% have strong inhibition (average inhibition zone diameter of 12.95 mm to 20.75 mm). Concentrations of 40% and 20% show moderate inhibition (8.6 mm and 6.8 mm). The positive control (gentamicin) gives the highest inhibition zone (20.75 mm), while the negative control (DMSO 20%) does not show an inhibition zone. Conclusion: Testing with the Kirby-Bauer method revealed that red rose petal extract at concentrations of 100%, 80%, and 60% demonstrated strong inhibitory power, while concentrations of 40% and 20% were categorized as having moderate inhibitory power. The results showed that red rose petal extract was proven to be effective in inhibiting the growth of S. aureus.
