Abstract
Pada tahun 2018, 74,1% masyarakat Indonesia mempunyai masalah pada kesehatan gigi dan mulut termasuk karies gigi dan penyakit periodontal. Infeksi pada rongga mulut disebabkan oleh berbagai penyebab, salah satunya adalah Enterococcus faecalis. Enterococcus faecalis merupakan bakteri Gram positif yang dapat menyebabkan kegagalan perawatan saluran akar gigi. Biji kopi mengandung berbagai senyawa metabolit sekunder antara lain kafein, trigonelin, asam klorogenat, polifenol dan flavonoid yang bersifat antibakteri. Penelitian ini bertujuan untuk mengetahui efektivitas antibakteri dari ekstrak ethanol 96% Biji Hijau Kopi Robusta (Coffea canephora) dalam menghambat pertumbuhan bakteri Enterococcus faecalis. Penelitian ini adalah eksperimental laboratorik dengan metode Kirby-baurer disk diffusion. Konsentrasi ekstrak Biji Hijau Kopi Robusta yang digunakan adalah 100 mg/ml, 50 mg/ml, 25 mg/ml, 12,5 mg/ml, 6,25 mg/ml, 3,125 mg/ml, dan 1,5625 mg/ml aquadest. Sebagai kontrol positif digunakan Ampicilin, dan aquadest sebagai kontrol negatif. Rataan diameter zona hambat bakteri Enterococcus faecalis adalah 26,08 mm dan 0 mm untuk masing masing kontrol positif dan kontrol negatif. Rataan diameter zona hambat bakteri terhadap ekstrak Biji Hijau Kopi Robusta secara berurutan adalah 14,75 mm, 10,5 mm, 6,15 mm, 5,7 mm, 4,4 mm, 2,9 mm, dan 1,8 mm. Infections in the oral cavity are caused by various causes, one of which is Enterococcus faecalis. Enterococcus faecalis is a Gram-positive bacteria that can cause root canal treatment failure. Coffee beans contain various secondary metabolite compounds including caffeine, trigonelline, chlorogenic acid, polyphenols, and flavonoids, which are antibacterial. This study aims to determine the antibacterial effectiveness of 96% ethanol extract of Robusta Green Coffee Beans (Coffea canephora) in inhibiting the growth of Enterococcus faecalis bacteria. This research is a laboratory experimental with the Kirby-Baurer disk diffusion method. The concentration of Robusta Coffee Green Bean extract used was 100 mg / ml, 50 mg / ml, 25 mg / ml, 12.5 mg / ml, 6.25 mg / ml, 3.125 mg / ml, and 1.5625 mg / ml. aqua dest. Ampicillin was used as a positive control and aqua dest as a negative control. The mean diameter of the inhibition zone for Enterococcus faecalis bacteria was 26.08 mm and 0 mm for positive and negative controls, respectively. The average diameter of the bacterial inhibition zone for Robusta Green Coffee Beans extract was 14.75 mm, 10.5 mm, 6.15 mm, 5.7 mm, 4.4 mm, 2.9 mm, and 1.8 mm, respectively.
