Abstract

Indonesia mempunyai sumber daya alam yang sangat berlimpah seperti keanekaragaman pada biota laut, salah satunya adalah balakutak (Sepia officinalis). Balakutak memiliki potensi sebagai sumber gizi karena kandungan omega 3, kandungan protein, lemak, mineral, antioksidan dan yang lainnya. Hasil uji fitokimia pada daging balakutak dengan ekstrak etanol ditemukan beberapa senyawa alkaloid, triterpenoid, tanin dan glikosida. Saat ini, sebagian besar masyarakat hanya mengkonsumsi balakutak sebagai lauk saja. Padahal, daging balakutak memiliki kemungkinan untuk diolah menjadi sumber antioksidan. Penelitian terkait manfaat dan kandungan senyawa kimia dari tinta sudah banyak dilakukan. Namun, penelitian terkait daging balakutak masih jarang dilakukan. Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan dari ekstrak daging balakutak. Penelitian ini bersifat eksperimental in vitro yang bertujuan untuk menguji aktivitas antiradikal bebas DPPH pada berbagai tingkatan konsentrasi (10ppm, 50ppm, 100ppm, 150ppm, dan 200ppm). Dilakukan maserasi selama 24 jam dengan 3 kali pengulangan dan dilakukan proses evaporasi menggunakan rotary vacuum evaporator. Pada uji aktivitas antioksidan dengan menggunakan metode DPPH (2,2 Difenil -1- Pikrilhidrazil) didapat hasil persentase daya hambat terbesar pada 47,79%. Berdasarkan data yang didapat disimpulkan bahwa ekstrak daging balakutak dengan pelarut etanol memiliki tingkat antioksidan yang sangat lemah. Kata kunci: antioksidan, balakutak, DPPH, radikal bebas. / Indonesia has abundant natural resources such as diversity in marine biota, one of which is the balakutak (Sepia officinalis). Balakutak has the potential as a source of nutrition because of the omega 3, protein, fat, minerals, antioxidants, and others. Phytochemical test results on balakutak flesh with ethanol extract found several alkaloids, triterpenoids, tannins and glycosides. Currently, most people only consume balakutak as a side dish. Whereas balakutak has a potential of becoming antioxidant source. Many studies have been carried out regarding the benefits and content of chemical compounds from the ink. However, research on balakutak flesh is still rare. This study aims to determine the antioxidant activity of balakutak flesh extract. This study was an in vitro experimental study aimed at testing the free anti-radical activity of DPPH at various concentration levels (10ppm, 50ppm, 100ppm, 150ppm, and 200ppm). Maceration was carried out for 24 hours with 3 repetitions and the evaporation process was carried out using a rotary vacuum evaporator. In the antioxidant activity test using the DPPH method (2,2 Diphenyl -1-Picrylhydrazyl) the highest percentage of inhibition obtained was 47.79%. Based on the data obtained it was concluded that balakutak flesh extract with ethanol solvent has a very weak antioxidant level. Keywords: antioxidant, balakutak, DPPH, free radical
