Abstract

Antioksidan ialah zat yang dapat meredakan radikal bebas yang sudah terbentuk didalam tubuh misalnya, penuaan kulit. Beras hitam merupakan salah satu spesies Oryza sativa L. yang memiliki pigmen berupa aleuron. Pigmen antosianin pada beras hitam berperan sebagai antioksidan. Penelitian yang dilakukan untuk mengetahui kekuatan aktivitas antioksidan dan kandungan metabolit sekunder beras hitam. Aktivitas antioksidan dilakukan dengan metode 2,2-diphenyl-1- picrylhydrazyl (DPPH) dan ferric reducing antioxidant power (FRAP), sedangkan skrining fitokimia dilakukan dengan metode Harbone. Ekstrak beras hitam dimaserasi dengan pelarut etanol 96% pro-analisis (p.a). Berdasarkan hasil penelitian bahwa pengujian aktivitas antioksidan dengan teknik DPPH menunjukkan ekstrak beras hitam memiliki aktivitas antioksidan kuat dengan nilai Inhibition Concentration 50% (IC50) sebesar 97,34 ppm, sedangkan pengujian aktivitas antioksidan menggunakan metode FRAP menunjukkan ekstrak beras hitam memiliki aktivitas antioksidan sedang dengan nilai IC50 sebesar 181,83 ppm. Hasil skrining fitokimia menunjukkan positif flavonoid dan steroid. Kata kunci : Antioksidan, DPPH, Fitokimia, FRAP, Oryza sativa L.indica. / Antioxidants are substances that can relieve free radicals that have formed in the body, for example, skin aging. Black rice is one of the species of Oryza sativa L. which has a pigment in the form of aleuron. Anthocyanin pigments in black rice act as antioxidants. Research conducted to determine the strength of antioxidant activity and the content ofsecondary metabolites of black rice. Antioxidant activity is carried out by the method of 2,2-diphenyl-1-picrylhydrazyl (DPPH) and ferric reducing antioxidant power (FRAP), while phytochemical screening is carried out by the Harbone method. Black rice extract macerated with 96% pro-analysis (p.a) ethanol solvent. Based on the results of the study that antioxidant activity testing with the DPPH technique showed black rice extract has strong antioxidant activity with an Inhibition Concentration value of 50% (IC50) of 97.34 ppm, while antioxidant activity testing using the FRAP method showed black rice extract has moderate antioxidant activity with an IC50 value of 181.83 ppm.
