Abstract

Kanker payudara masih menduduki peringkat pertama sebagai kanker yang memiliki tingkat mortalitas tinggi pada wanita. Timbulnya efek samping dan kemo resistensi mendorong pengembangan terapi alternatif dari bahan herbal. Jintan hitam diketahui mengandung senyawa-senyawa yang memiliki aktivitas antioksidan dan sitotoksik terhadap sel kanker. Penelitian ini bertujuan untuk mengetahui potensi ekstrak etanol jintan hitam (Nigella sativa L.) sebagai agen kemoterapi melalui inhibitory concentration (IC50) pada model sel kanker payudara tipe estrogen reseptor positif MCF-7. Jintan hitam (Nigella sativa L.) diekstraksi dengan etanol 96% dengan metode maserasi dan karakterisasi senyawa aktif onedengan uji fitokimia secara kualitatif. Pada proses identifikasi dengan uji tabung bahwa hasil uji fitokimia menunjukkan jintan hitam mengandung golongan senyawa alkaloid, tanin, saponin, flavonoid, triterpenoid, dan steroid. Uji sitotoksik dilakukan pada kanker payudara tipe estrogen reseptor positif MCF-7 menggunakan metode MTT- assay dengan berbagai konsentrasi. Menurut NCI (National Cancer Institute) suatu ekstrak dikatakan aktif jika (IC50 <30 μg/mL), moderat aktif (30 μg/mL ≤ IC50 100 μg/mL), tidak aktif (IC50 >100 μg/mL). Analisis hasil dilihat dari kemampuan ekstrak yang mampu membunuh 50% sel kanker (IC50) berdasarkan hasil pembacaan absorbansi gelombang 570 nm pada ELISA reader. Nilai IC50 diperoleh dari kurva regresi linear dengan menghubungkan nilai logaritma konsentrasi ekstrak sampel sebagai X dan nilai presentasi kematian sel sebagai Y (% inhibisi). Ekstrak uji yang digunakan dengan berbagai konsentrasi 500 μg/mL; 250 μg/mL; 62,5 μg/mL; 15,625 μg/mL, 7,8125 μg/mL. Ekstrak etanol 96% jintan hitam memiliki aktivitas sitotoksik terhadap sel kanker payudara tipe estrogen reseptor positif MCF-7 dengan nilai IC50 sebesar 62.859 μg/mL (kategori moderat aktif). Hasil uji korelasi dan regresi menunjukan bahwa konsentrasi ekstrak jintan hitam mempengaruhi % inhibisi sel MCF-7 secara signifikan (p < 0.05). Hal ini mengindikasikan jintan hitam memiliki potensi untuk dikembangkan sebagai terapi antikanker di masa depan. Kata kunci : Jintan Hitam, MCF-7 ER (+), Fitokimia, Sitotoksik. / Breast cancer remains the malignancy with the highest mortality rate among women. The emergence of adverse effects and chemotherapy resistance encourages the development of herbal-based alternatives. Black cumin is known to contain compounds with anti-cancer antioxidant and cytotoxic properties. This study aim to evaluate the potential of ethanol extract of black cumin (Nigella sativa L.) as a chemotherapeutic agent by determining the inhibitory concentration (IC50) in an MCF-7 breast cancer cell model with an estrogen receptor that is positive. Black cumin (Nigella sativa L.) was extracted with 96% ethanol using the maceration technique, and the active compounds were characterized using qualitative phytochemical assays. The results of the phytochemical test revealed that black cumin contains alkaloids, tannins, saponins, flavonoids, triterpenoids, and steroids during the test tube identification procedure. Using the MTT-assay method and varying concentrations, a cytotoxic test was performed on MCF-7 estrogen receptor-positive breast cancer using the MTT-assay method. According to the National Cancer Institute (NCI), an extract is active if (IC50 <30 μg/mL), moderately active if (30 μg/mL ≤ IC50 100 μg/mL), and inactive if (IC50 >100 μg/mL). Based on the results of reading the absorbance of the 570 nm wave on the ELISA reader, the ability of the extract to eliminate 50% of cancer cells (IC50) served as the basis for the analysis of the results. The IC50 value was derived from the linear regression curve by correlating the logarithmic value of the sample extract concentration (X) with the percentage of inhibition (Y) (% inhibition). Various concentrations of 500 μg/mL; 250 μg/mL; 62,5 μg/mL; 15,625 μg/mL, 7,8125 μg/mL were applied to test extracts. With an IC50 value of 62,859 μg/mL against estrogen receptor-positive MCF-7 breast cancer cells, the 96% ethanol extract of black cumin is moderately cytotoxic. The results of correlation and regression tests showed that the concentration of black cumin extract significantly affected the % inhibition of MCF-7 cells (p <0.05).
