Abstract
Pseudomonas aeruginosa merupakan bakteri Gram negatif yang mampu menyebabkan infeksi kulit seperti dermatitis, folikulitis, infeksi pada luka bakar, hingga infeksi pada ulkus diabetikum. Penggunaan antibiotik secara irasional akan membuat angka resistensi meningkat sehingga infeksi sulit disembuhkan. Untuk itu perlu dilakukan upaya pencarian antibakteri alternatif. Tujuan penelitian ini adalah untuk mengetahui aktivitas antibakteri ekstrak beras hitam varietas cempo ireng (Oryza sativa L. indica) terhadap bakteri Pseudomonas aeruginosa. Beras hitam diekstraksi secara maserasi menggunakan pelarut etanol 96% dan kloroform 99.8% dan diuji aktivitas antibakterinya menggunakan metode Kirby-Bauer disk diffusion dengan konsentrasi ekstrak beras hitam 100 mg/ml, 50 mg/ml, 25 mg/ml, 12,5 mg/ml, dan 6,25 mg/ml. Kontrol positif menggunakan antibiotik Gentamisin dan kontrol negatif menggunakan DMSO. Pada penelitian ini tidak tampak zona hambatan di sekitar cakram uji yang mengandung ekstrak beras hitam dengan berbagai konsentrasi. Berdasarkan hasil penelitian tersebut dapat disimpulkan bahwa ekstrak beras hitam varietas cempo ireng (Oryza sativa L. indica) tidak terbukti efektif dalam menghambat pertumbuhan bakteri Pseudomonas aeruginosa. / Pseudomonas aeruginosa is a Gram-negative bacteria that can cause skin infections such as dermatitis, folliculitis, infections in burn wounds, and infections in diabetic ulcers. Irrational use of antibiotics will increase the resistance rate, making infections difficult to cure. For this reason, efforts need to be made to search for alternative antibacterials. The aim of this research was to determine the antibacterial activity of black rice extract of the cempo ireng variety (Oryza sativa L. indica) against Pseudomonas aeruginosa bacteria. Black rice was extracted by maceration using 96% ethanol and 99.8% chloroform solvents and tested for antibacterial activity using the Kirby-Bauer disk diffusion method with black rice extract concentrations of 100 mg/ml, 50 mg/ml, 25 mg/ml, 12.5 mg/ml. ml, and 6.25 mg/ml. The positive control uses the antibiotic Gentamicin and the negative control uses DMSO. In this study, no inhibition zone appeared around the test disks containing black rice extract at various concentrations. Based on the results of this research, it can be concluded that the black rice extract of the cempo ireng variety (Oryza sativa L. indica) has not proven effective in inhibiting the growth of Pseudomonas aeruginosa bacteria.
