Abstract

Latar Belakang: Indonesia memiliki kekayaan sumber daya alam yang sangat berlimpah seperti keanekaragaman pada biota laut, salah satunya adalah tulang Balakutak (Sepia officinalis). Balakutak mempunyai potensi sebagai sumber gizi karena kandungan omega 3 didalamnya. Selain itu, penelitian juga menunjukan adanya kandungan protein, lemak, mineral, antioksidan dan yang lainnya. Hasil uji fitokimia pada tulang balakutak dengan ekstrak etanol ditemukan beberapa senyawa alkaloid, steroid, triterpenoid, tanin dan glikosida. Saat ini, sebagian besar masyarakat yang mengkonsumsi balakutak akan membuang tulang yang terdapat dibagian tubuhnya. Padahal, tulang balakutak bisa diolah menjadi berbagai macam kebutuhan. Penelitian terkait manfaat dan kandungan senyawa kimia dari kepala, badan, tinta, dll sudah banyak dilakukan. Namun, penelitian tentang tulang balakutak masih jarang dilakukan. Tujuan Penelitian: Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan dari ekstrak tulang balakutak. Metode Penelitian: Penelitian ini bersifat eksperimental in vitro yang bertujuan untuk menguji aktivitas antiradikal bebas DPPH pada berbagai tingkatan konsentrasi (10ppm, 50ppm, 100ppm, 150ppm, dan 200ppm). Dilakukan maserasi selama 24 jam dengan 3 kali pengulangan dan dilakukan proses evaporasi menggunakan rotary vacuum evaporator. Hasil Penelitian: Pada uji aktivitas antioksidan dengan menggunakan metode DPPH (2,2-Difenil-1-Pikrilhidrazil) didapat hasil persentase daya hambat terbesar pada 48,42%. Kesimpulan: Berdasarkan data yang didapat disimpulkan bahwa ekstrak tulang balakutak dengan pelarut etanol memiliki tingkat antioksidan yang sangat lemah. Kata kunci: antioksidan, radikal bebas, DPPH, balakutak. / Background: Indonesia has abundant natural resources such as diversity in marine biota, one of which is the Balakutak bone (Sepia officinalis). Balakutak has the potential as a source of nutrition because of the omega 3 content in it. In addition, research also shows the presence of protein, fat, minerals, antioxidants and others. Phytochemical test results on balakutak bone with ethanol extract found several alkaloids, steroids, triterpenoids, tannins and glycosides. Currently, most people who consume balakutak will throw away the bones in their body parts. In fact, balakutak bone can be processed into various kinds of needs. Many studies have been carried out regarding the benefits and content of chemical compounds from the head, body, ink, etc. However, research on balakutak bones is still rare. Research Objectives: This study aims to determine the antioxidant activity of balakutak bone extract. Research Methods: This study was an in vitro experimental study aimed at testing the free anti-radical activity of DPPH at various concentration levels (10ppm, 50ppm, 100ppm, 150ppm, and 200ppm). Maceration was carried out for 24 hours with 3 repetitions and the evaporation process was carried out using a rotary vacuum evaporator. Research Results: In the antioxidant activity test using the DPPH method (2,2-Diphenyl-1-Pikrylhidrazil) the highest percentage of inhibition was obtained at 48.42%. Conclusion: Based on the data obtained it was concluded that balakutak bone extract with ethanol solvent has a very weak antioxidant level. Keywords: antioxidant, free radical, DPPH, balakutak
